Place your plexiglass sheet on top of a piece of scrap plexiglass one that is already ruined or a piece of medium density fiberboard mdf this will make it less likely that you will chip the back of the board or scratch it when the drill bit goes through. Depress the trigger of the drill motor to start drilling the hole into the acrylic sheet. Line the plywood with a rubber pad sheets of newsprint or some type of cushioning material. The hole saw works best at low speeds 100 to 200 rpm.

Then insert a diamond drill bit into a drill and position it over the spot you want the hole to be in. While you are drilling through the plexiglass sheet the friction would produce intense heat and the bits would cause lots of plastic shavings around the hole especially if the sheet is thick. Step 6 drill through the acrylic sheet and into the 3 8 inch plywood.
